A fully funded Postdoc position is availablein the lab Structural & Molecular Microbiology at VIB VrijeUniversiteit Brussel, Belgium, to work on an FWO project studyingthe nucleation, cross-reactivity, and structure of bacterialfunctional amyloids (FA).

Amyloids areaggregative protein or peptide fibrils best known for theirimplication in a range of neurodegenerative illnesses. Theself-assembly into cross- -structured fibrils is however notrestricted to the off-pathway protein misfolding events, but alsofound as the native conformation of a wide range of pro- andeukaryotic proteins referred to as FA. FAs produced by prokaryotesare often related to the persistence and virulence of bacterialbiofilms. Despite their importance, little is known regarding theFA ultrastructure and their mechanism of formation.

In this project we seek to determine the fiberstructures and nucleation mechanisms of 3 selected FAs thatcontribute to the virulence of well-known human pathogens. Becausethere is an increasing awareness that bacterial FAs can trigger theco-aggregation of pathologic amyloids, we will also study thecross-reactivity between our FA targets and two human diseaseamyloid model systems.

We aim to deepen theunderstanding of how bacteria handle these potentially cytotoxicproteins, inspire novel antibacterial strategies by targeting FAvirulence factors and expose additional health risk factorsassociated with FA triggered amyloidosis of host proteins.

The tasks will be production and purification ofthe amyloid proteins, biophysical characterization of theiramyloidogenicity, determining cross-reactivity with pathologicalamyloids, and determining the fibril structures using cryo electronmicroscopy.

Profile

Candidates should hold a PhD in Sciences (eg.Biochemistry, Chemistry, Physics or Biophysics) or Bio-engineeringsciences. Candidates should have a strong background in recombinantexpression and purification of proteins, and have a keen interestin molecular assembly and structural biology.
